  1. The major scientific directions: Genomic selection of crops in Kazakhstan, DNA-based genotyping of wild flora in Kazakhstan.
  2. The main results of work (within the last 3 years):

Studying the genetic diversity of domestic cultivars and promising lines of wheat, barley, rice, oat, soybean, chickpea, and pea using microsatellite DNA markers.

Genotyping of cultivars collections and lines of wheat, barley, rice and soybean based on a new genomic technology using Illumina SNP-markers.

Studying problems of wheat, barley and soybean adaptations using specific flowering-genes that control the vernalization processes and the response to photoperiod length.

Studying the genetics of the barley collection’s resistance to brown spot disease and stem rust.

Identification of DNA-markers for quantitative trait loci associated with the wheat quality.

Creation of the DNA-bank for endemic, rare and endangered species of flora in Kazakhstan.

Creation of two spring barley cultivars: Shakhristan (2013, in cooperation with Kyzylorda Research Institute of Rice breeding) and Bozman (2014, in cooperation with Karaganda Research Institute of Plant breeding and selection) together with Kazakhstan breeders.

Scientific-pedagogical activity:

 

Turuspekov Y. is a part-time lecturer (0.25 FTE) in al-Farabi Kazakh National University, where he reads lectures on «Genomics» for masters and PhD students in the Department of Biotechnology, Faculty of Biology and Biotechnology.

 

Abugalieva S. is a co-supervisor of studies of PhD student  Zatybekov A. (KazNAU).

 

Turuspekov Y. is a co- supervisor of studies of PhD student Ermekbayev K. (John Innes Centre, UK, “Bolashak” program).

 

There are 3 master degree research works of al-Farabi Kazakh National University students under the supervison of Turuspekov Y. and Abugalieva S.

 

Turuspekov Y. is a member of the Dissertation Council on thesis defense in the field of biology, ecology and biotechnology at al-Farabi Kazakh National University.

Introduction to the problem. Safety instructions.  Work process in the lab. Issuance of the course materials.

Day 1Methods of express DNA extraction for PCR. Preparation of solutions. Disruption of cells and tissues, removing of proteins and impurities. DNA Precipitation.

Day 2Checking the quality and quantity of DNA. Agarose gel electrophoresis. Spectrophotometer.

Day 3. PCR theory. Formulation of the problem. Ingredients of the  reaction.  PCR running.

Day 4. Gels filling. Electrophoresis. Gel — documentation system. Documenting of results. Cutting off of DNA-bands from agarose gels.

Day 5Using of the commercial kits for DNA purification.

Day 6PCR for DNA sequencing. Purification of PCR products from the excess of dye, primers and other impurities.

Day 7Introduction to the DNA sequencing machine (DNA sequencer). Start of DNA sequencing.

Day 8Documentation of the sequencing results. Comparative analysis of nucleotide sequences.
